Abstract
This study focused on the isolation, identification, and preservation of yeast from natural sources within Nasarawa Town, Nasarawa State, Nigeria. Samples were collected from fruits and flower nectar and cultured using standard microbiological techniques on Potato Dextrose Agar (PDA). Distinct yeast colonies obtained were purified and identified through cultural, microscopic, and biochemical characterization. The isolates exhibited cream-to-white coloured colonies with smooth and dome-shaped appearances, while microscopic examination revealed round to oval budding yeast cells. Biochemical tests showed that the isolates were capable of fermenting glucose, sucrose, and maltose. Based on the observed characteristics, the isolates were presumptively identified as Candida glabrata, Candida albicans, and Saccharomyces cerevisiae. The study further demonstrated that fruits and flower nectar are suitable natural habitats for indigenous yeasts with potential industrial and biotechnological applications. Preservation methods employed successfully maintained the viability of the isolates for future use.
